Test Of R.B.C Blood Count
AIM :- Enumerate the total R.B.C count of your own Blood
APPARATUS :-
Haemocytometer, sterilized pricking needle, R.B.C Pipette , Compound Microscope, Hayem diluting tube
PRINCIPLE :-
Haemocytometer is a kind os slide containing counting chamber, the (Neubaur) or any other cytometer may be used, the neubaur cytometer may be used , the neubaur cytometer consist double cell slide with 2 sunk platform in open cell type . each platform has a ruling so that replicate duplicate count may br made , when special cover glass kept in position the depth of 10mm is maintain over the ruling
, after ward diluted blood is run over after cover glass has been fixed
Note :-
Counting chamber are 9 large sqaure having sides of 1 mm each , Centre square with A,B,C,D is subdivided into 25 medium sqaure , a each medium intern contain 16 tiny square having sides of 0.05 mm long
CALCULATION :-
Number of R.B.C /cubic mm = Number of cell counted >< Dilution >< 400 / Number of small square counted = 1115 >< 200 >< 400 / 80 = 1,115,000 / cubic mm => 1.11 million / mm(cube)
